That's a very broad subject.
I suggest you check out some of the sites set up specifically to assist webmasters, ecommerce operators and/or online retailers.
In any event, you'll need to be more specific about what you intend to sell, how you're going to acquire stock, how you intend to fulfil orders, returns policies, warranties, etc. and the minutiae of running a retail business, before you fill in the relatively small blank that is the online stuff...

To build a site with no knowledge of HTML, buy Frontpage 2003 or Expression Web (slightly more involving to Frontpage)
A good web host would be www.1and1.co.uk.
If you intend to sell e-goods I recommend you use Paypal or WorldPay to handle payments.
If you wish to send the goods securely after payment, try DLGuard which integrates to both Paypal and WorldPay.
For a software installer I would Install Creator. To create updates I would use Patch Maker.
To market your product and achieve instant Search Engine presence, use Google Adwords.
To create revenue for your site, try Google Adsense
Also ensure your site has been reviewed by Site Advisor.
